Literature Sharing | Molecular mechanism of SmMYB53 activates the expression of SmCYP71D375, thereby modulating tanshinone accumulation in Salvia miltiorrhiza

This study uncovers the transcriptional regulatory network controlling SmCYP71D375, a key enzyme involved in tanshinone biosynthesis in Salvia miltiorrhiza. Using the promoter of SmCYP71D375 as bait in a yeast one-hybrid screen, researchers identified SmMYB53, an R2R3-MYB transcription factor, as an upstream regulator. Overexpression of SmMYB53 in transgenic hairy roots increased SmCYP71D375 expression and promoted tanshinone accumulation, while RNAi-mediated silencing of SmMYB53 reduced it.

Porcine reproductive and respiratory syndrome virus (PRRSV) is a single-stranded positive-sense RNA virus with an envelope. It-encoded non-structural protein 1α (nsp1α) plays a key role in evading host immune responses. Exploring the interaction between host factors and PRRSV nsp1α is crucial for understanding the mechanism of virus immune escape and virus control. Here, we constructed a cDNA library using porcine lung tissues and identified 33 potential host proteins interacting with viral nsp1α using yeast two-hybrid (Y2H) screening. These interactions were further analyzed using Gene Ontology and KEGG pathway analysis.

A working model was proposed in which LoMYB26 interacts with LoJAZ4 to participate in JA-mediated endothecium lignification and anther dehiscence. When bioactive JA (JA-Ile, Me-JA) accumulates, LoJAZ4 is degraded, releasing the transcription factor LoMYB26. LoMYB26 directly binds to the LoCOMT promoter and activates its expression to promote lignin deposition in the anther endothecium. The secondary wall thickening of the anther endothecium results in timely anther dehiscence.
