In this assay, two proteins with different affinity tags (e.g., GST, His, or Flag) are expressed and purified separately, then incubated together under defined conditions. One of the proteins is immobilized via its tag on an affinity matrix, and interacting partners are pulled down and detected. If protein A pulls down protein B, it indicates a direct interaction between them.
