When a functional signal peptide is fused to SUC2, the invertase secretion function is restored, allowing the YTK12 strain to grow on selective media with raffinose as the carbon source.

    To verify the signal peptide secretion functionality, a TTC (triphenyltetrazolium chloride) colorimetric reaction is used. This simple and reliable method allows researchers to confirm the success of their signal peptide fusions and optimize their yeast expression systems for downstream applications.
