This kit uses the YTK12 yeast strain, which lacks the invertase gene and cannot grow on media where sucrose is the sole carbon source. The included pSUC2 plasmid contains a tryptophan synthesis gene and an invertase gene (SUC2) missing its signal peptide and the start codon (ATG). When a functional signal peptide is fused to SUC2, the invertase secretion function is restored, allowing the YTK12 strain to grow on selective media with raffinose as the carbon source.
To verify the signal peptide secretion functionality, a TTC (triphenyltetrazolium chloride) colorimetric reaction is used. This simple and reliable method allows researchers to confirm the success of their signal peptide fusions and optimize their yeast expression systems for downstream applications.
