Workflow Overview
From Colony to Gel in 3 Simple Steps
· Yeast Cell Lysis: Use Yeast Buster Pro to disrupt cells at 100°C for 5 min.
· PCR Reaction: Amplify target gene with optimized primers and ready-to-use 2× PCR mix.
· Gel Electrophoresis: Load PCR product directly onto gel for analysis.
Experimental Protocol Summary
Lysis Reaction Setup:
Add 5 μl Yeast Buster Pro to PCR tube
Pick colony and suspend in lysis buffer
Heat at 100°C for 5 min
PCR Reaction Setup (Total volume: 50 μl):
25 μl 2× Yeast PCR Mix
2 μl Forward Primer (10 μM)
2 μl Reverse Primer (10 μM)
1 μl lysate as template
Add ddH₂O to 50 μl
Thermal Cycler Conditions:
Initial Denaturation: 95°C, 3 min
30–32 cycles of:
Denaturation: 95°C, 15 sec
Annealing: Tm − 5°C, 15 sec
Extension: 72°C, 15 sec/kb
Final Extension: 72°C, 5 min
Hold: 4°C
