Maize Transcription Factor Library
Maize Transcription Factor Library for Efficient Gene Screening and Interaction Studies
Service Overview
The Maize Transcription Factor Library is a comprehensive collection featuring a wide array of transcription factors from Zea mays—commonly known as maize or corn. This library includes open reading frame (ORF) clones of transcription factors, obtained using advanced genetic and molecular biology techniques. The transcription factor genes are readily available for direct incorporation into vectors, streamlining and enhancing the efficiency of downstream screening applications such as Yeast One-Hybrid (Y1H) and Yeast Two-Hybrid (Y2H) assays.
Key Applications of the Maize TF Library
Gene Function Studies: Identify key transcription factors regulating maize gene expression.
Transcription Factor Screening: Efficiently screen for transcription factors that interact with target genes.
Crop Improvement Research: Explore transcription factors related to stress resistance, yield, and other agronomic traits in maize.
How Y1H and Y2H Screening with Maize TF Library Works
The Maize Transcription Factor Library is versatile, designed for both Y1H and Y2H screening assays. These assays enable the identification and study of transcription factors that regulate gene promoters and interact with other proteins.
Yeast One-Hybrid Screening (Y1H): The yeast GAL4 protein acts as a transcription factor. The DNA-binding domain, located near the C-terminal, activates the yeast β-galactosidase gene. By fusing the GAL4 DNA-binding domain to your target gene, you can efficiently screen for transcription factors that interact with your promoter of interest.

Yeast Two-Hybrid Screening (Y2H): The Gal4 transcription factor consists of a DNA-binding domain (BD) and a transcription activation domain (AD). These domains can be independently expressed and retain functionality. The Y2H assay allows you to screen for interacting partners by fusing your target protein to the BD and screening for interactions with transcription factors from the library.

Technical Advantages
Full-Length cDNA Screening: Avoid frame shift issues with full-length cDNA clones.
Direct Downstream Application: Results from screening can be directly used in downstream experiments, eliminating the need for one-to-one verification.
Broad Compatibility: Suitable for both Y1H and Y2H screening assays.
Comprehensive Library: Includes over 1,900 full-length cDNA clones from 81 transcription factor families, providing a broad and deep resource for transcription factor screening.
