1. Why is the library length of random sequences only 7bp?
Normally, DNA motifs are fairly short (5-20 base pairs [bp] in length). The motif length ranges from 2 to 10 bases. The graphic presentation and analysis of the motif distribution found that the motif length range is mainly concentrated from 2 to 10 bases.
Considering the workload and cost of new element’s boundary determination, we recommend 7bp;
2. Why is the random sequence inserted into the SmaI digestion site?
Because this enzymatic sequence is CCCGG, this sequence can be used exactly as a marker site to identify the inserted sequence, that is, it forms CCCHNNNNNNCGGG with CCC and GGG as marker sites.
