Bait and Prey Yeast Two Hybrid Assay is a molecular biology technique used to study protein interactions and is one of the important tools for analyzing protein interactions. Yeast two-hybrid experiments are mainly used to identify proteins that interact within cells and to understand the structural and functional relationships between them. The basic principle of Bait and Prey Yeast Two Hybrid Assay is to utilize the natural physiological characteristics of yeast cells. Transcription factors in yeast cells are divided into two parts, one is the DNA-binding domain and the other is the transactivation domain. These two parts are each attached to the two proteins to be studied. If the two proteins interact within the cell, the two parts of the transcription factor recombine, activating a reporter gene that produces an observable phenotype.
Bait and Prey Yeast Two-Hybrid Assay consists of two main steps. First, the coding sequence of the protein to be tested is connected to the DNA binding domain and transactivation domain respectively, and inserted into yeast cells. The coding sequence for another protein is then linked to another part of the transcription factor and inserted into the yeast cell as well. If the two proteins in the Bait and Prey Yeast Two Hybrid Assay interact, the two parts of the transcription factor will combine, activating the reporter gene, ultimately leading to the appearance of the phenotype.
